PriCells: Recombinant Human Thrombopoietin (rhTPO)
Thrombopoietin (TPO), the c-Mpl ligand, is the primary physiologic regulator of megakaryocyte and platelet development. TPO is synthesized primarily in the liver as a single 353-amino acid precursor protein. On removal of the 21-amino acid signal peptide, the mature molecule consists of 2 domains: a receptor-binding domain that shows considerable homology to erythropoietin and a carbohydrate-rich carboxy-terminus of the protein that is highly glycosylated and important in maintaining protein stability. TPO in treating chronic nonchemotherapy-induced thrombocytopenia associated with myelodysplastic syndromes, idiopathic thrombocytopenic purpura, thrombocytopenia due to human immunodeficiency virus, and liver disease. TPO is effective in reducing surgical thrombocytopenia and bleeding, ex vivo expansion of pluripotent stem cells, and as a radioprotectant.
