PriCells: Recombinant Human Granulocyte-Macrophage Colony Stimulating
Granulocyte-macrophage colony-stimulating factor, often abbreviated to GM-CSF, is a protein secreted by macrophages, T cells, mast cells, endothelial cells, and fibroblasts. GM-CSF contains N-linked and possibly O-linked oligosaccharides. This compares with unmodified GM-CSF that has a predicted molecular mass of 14.5 kDa. Purified GM-CSF consists of 5-50% carbohydrate by weight. Production: rhGM-CSF is produced in E. coli and purified via sequential chromatography. Filtered prior to lyophilization through a 0.22 micron sterile filter. Biological Activity: ED50 range = 0.02 to 0.1 ng/mL, determined by the dose dependent proliferation of human TF-1 cells. Optimal concentration for individual application should be determined by a dose response assay. Negative test: HIV-1, HBV, HCV, Mycoplasmas, Bacteria, yeast, and fungus Protein Form: recombinant human Expression and Purification: rhGM-CSF is expressed in E. coli and purified via sequential chromatography. Contents and Storage: Store this lyophilized preparation at -20°C, preferably desiccated. Upon reconstitution, apportioninto working aliquots and store at -20°C. Avoid repeated freeze/thaw cycles.
